Picking up right after the events of the film, Megamind: Ultimate Showdown lets players assume the role of Megamind in order to take on the villains of the Doom Syndicate. Megamind alternates between four weapons - a De-Stroy gun (pew pew pew), a De-Hydrate gun (shrinks objects and enemies), a blast gauntlet (watch your enemies struggle to stay upright as you blast them with your handheld tornado) and a Tesla Glove (creates shockingly powerful electrical discharges).

The game features around nine mission levels, with access to Megamind's Lair as a hub between missions. The player must collect glowing tokens known as BINKEYs in order to upgrade Megamind's weapons. BINKEY is collected by defeating Doom Syndicate enemies or destroying in-game items.

There are also unlocked mini-games throughout which can also be accessed from Megamind's Lair. Minigames include a Pod Racer, which is a simple timed race. The Sticky Boots game, which requires Megamind to climb up the side of a skyscraper using his magnetic boots while dodging obstacles thrown from the windows above. The Rampage game, a throwback to the classic arcade game Rampage. You are given the Megamind Battle Suit to destroy as many cities as possible within a given time. Mega-Blast game, another rampage style game but this time with a first person view from Megamind's Battle Suit. Finally the Mega-Ball game, where you'll find Megamind encased in a ball much like an 80s plasma globe. The goal is to control him and smash your way through an industrial-themed track before time runs out . Not easy, given the difficulty of maneuvering in what is essentially a hamster ball. The track will give the player the novelty feel of being inside a ping pong machine, with lots of twists and turns, moving environmental elements such as running through the moving truck trailers and pipes in which ones to drop to pull the player into a new Region.

Two-player co-op mode is included, although the second player doesn't have as much control as the Megamind character. It was designed to give a guardian or parent the ability to help their child, or younger sibling, without risking impeding the player's progress. Player two will control a brainbot with the ability to move around the level and shoot. All puzzles and tutorials are aided by Minion (the fish in the gorilla suit) who will ring the intercom whenever the player is obviously stuck with some helpful hints.
