Deal or No Deal is based on the game show of the same name. The show's host, Howie Mandell, also provides the tutorials for the game. The core gameplay follows the show, in that the player chooses one of twenty-six crates, containing an amount of money ranging from a hundred to a million dollars, then opens several more boxes. Eventually, a deal is offered by "The Banker", based on the average value of the remaining cases, which the player can either take or reject to open another batch of cases. Sometimes, after The Banker offers a deal, a mini-game may begin, giving the player a chance to win more money outside of the main game.

These mini-games, which can be played individually in "Arcade" mode (although in the DS version they must have been played first), include: Ice Breaker, in which the player must quickly remove ice to reach the frozen crate below; Shooter, in which the player must shoot boxes with positive monetary values and avoiding negative ones; Locker, in which the player must crack a six-digit code for a case; Balance, in which five crates must be weighed against each other, and the player wins money if their weights match; Memory, a simple memory matching game; Random, in which the player randomly chooses crates trying to avoid the empty crate (exclusively in the DS version); and Pyramid, in which cases with different values are shuffled and the player must choose the correct value (exclusively in the Wii version).

In the DS version, all the money the player earns can be spent in the "Showroom", with cosmetic unlocks such as cars and houses that can be upgraded with money. In the Wii version, it's only to build up a score.
